Okay my Gamesradar article topic has turned into what OUR predictions are so I decided to make a sperate page.
Now, I've learned some very interesting information after playing through the ending a second time.
Following the decoding of the codex pages, the group discovers that in order to open the (or as we now know, a) vault, two pieces of eden are required: One of the apples and the staff. During Minerva's message to Desmond, the final two slides she shows depict these two pieces of eden.
Actually being able to USE Pieces of Eden in Assassin's Creed III is like.. a must. What's the point of collecting all those shiny techno-magic things, without being able to press a Shoulder Button on your controller and using their abilities. Alternatively, they could be selected using the weapon wheel, but you can only have one equipped at a time. (You may need to go back to a Hideout of sorts to change weapons.)
Gamesradar has made their prediction( s ) for what AC3's plotline will be. It seems pretty accurate, but may not be to your liking, and may be completely wrong as well. They basically say that AC3 will become Mirror's Edge with an Assassins Creed twist on it, but I hope that's not the case. Anyway, here's the link, and there are predicted sequels for other games in there as well (but AC3 is at the top of the list):
